摘要:
目的评价中药复方养生茶对酒精所致肝损伤的保护作用及其机制,为该中药复方的开发和应用提供科学 资料。方法将60只雌性SD大鼠随机分为三个剂量组[0.83 g/(kg · bw)、1.67 g/(kg · bw)、5.00 g/(kg · bw) ], 以及空白组和模型组,连续30 d 按照2% (v/w) 容量灌胃,用50%乙醇以12 ml/(kg · bw)的剂量灌胃造成酒精 性急性肝损伤,全自动生化分析仪测定血清中天门冬氨酸氨基转移酶(AST)、丙氨酸氨基转移酶(ALT)、甘 油三酯(TG) 水平,生化法测定肝组织中氧化脂质(LPO)、超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶 (GSH)、丙二醛(MDA) 和甘油三酯(TG) 水平及病理检查。结果灌胃期间大鼠无不良反应。与模型组相 比,中药复方养生茶低剂量组ALT 水平降低(P<0.01),高剂量组GSH 水平有上升(P<0.05),三个剂量组的 LPO 浓度均下降(P<0.01),中剂量组(P<0.05)、高剂量组(P<0.01) MDA 水平降低,肝组织中脂滴含量降 低。结论中药复方养生茶可通过提高组织、细胞抗氧化能力,快速清除体内自由基,减轻其对肝组织的损 伤,减少血清脂质和肝脏脂质沉积,抑制脂类过氧化,从而缓解酒精性肝损伤。
Abstract:
Objective To evaluate the protective effects and probable mechanism of herbal compound health tea (HCHT) against alcohol liver damages of rat. Method Totally 60 female SD rats were randomly allocated to three HCHT groups [low dose group: 0.83g/ (kg · bw) , middle dose group: 1.67g/(kg · bw), high dose group: 5.00g/(kg · bw)], blank control group and model group. The HCHT groups were continuously received intragastric administration by 2% (v/w) for 30 days. Alcohol liver damages of rats were induced by intraperitoneal injection of alcohol with 12 ml/ (kg · bw) . Serum biochemical indexes (AST, ALT, TG) were detected by automatic biochemical analyzer. Several liver indexes [lipid peroxide (LPO) , malonyldialdehyde (MDA) , glutathione (GSH) , TG, superoxide dismutase (SOD) ] were examined, and pathological examination was carried out. Results Rats had no adverse reactions during gastric irrigation. Compared to the model control, ALT of low dose HCHT group was decreased (P<0.01) ; activity of GSH of high dose group was enhanced (P< 0.05) ; activities of LPO of three HCHT groups were decreased (P<0.01) ; MDA levels and content distribution of membranous lipid droplet of middle dose group and high dose groups were decreased (P<0.05 or P<0.01) . Conclusion This HCHT can improve cell antioxidant capacity, remove freeradicals, reduce the damage in liver, reduce serum lipid and liver lipid deposition, inhibition of lipid peroxidation, and thus relieve the alcoholic liver damage.
