摘要:
目的 观察不同浓度的仙鹤草水煎液对环磷酰胺(CP)致免疫抑制模型小鼠脾指数、白细胞含量的 影响。方法 BALB/C小鼠随机分为7组(正常对照组、模型对照组、治疗1组(仙鹤草10 g/ml浓 度)、治疗2组(仙鹤草5 g/ml浓度)、治疗3组(仙鹤草2.5 g/ml浓度)、治疗4组(仙鹤草1.25 g/ml浓 度)、治疗5组(仙鹤草0.625 g/ml浓度),每组15只。模型对照组和各治疗组小鼠腹腔注射CP(30 mg/kg) 制备免疫抑制模型,各组治疗每天给予不同浓度的仙鹤草水煎液灌胃(0.02 ml/g)。治疗5 d结束后,取 血(EDTA-K2抗凝)进行白细胞计数(WBC),摘取小鼠脾脏计算脾脏指数。实验数据采用SPSS13.0进 行统计分析,多组比较采用单因素方差分析,两两比较采用t检验,P<0.05表示差异有统计学意义。结果 与正常对照组比较,模型对照组脾脏指数、WBC极显著下降,差异有统计学意义(P<0.01)。与模型对照 组比较,治疗1组的WBC显著升高,差异有统计学意义(P<0.05),其余各治疗组的WBC均极显著升 高,差异有统计学意义(P<0.01)。各治疗组脾脏指数均显著高于模型对照组,差异有统计学意义(P< 0.01)。治疗3组、治疗4组、治疗5组脾脏指数、WBC数量明显高于正常对照组,差异有统计学意义 (P<0.01),且效果优于治疗1组和疗疗2组,差异有统计学意义(P<0.05)。结论 仙鹤草水煎液能显著 增强CP免疫抑制模型小鼠的免疫功能,随着给药浓度的下降各组小鼠免疫功能均明显提高,证明仙鹤草 的临床使用存在独特的量效关系。
Abstract:
Objective To evaluate how the water decoction of Agrimonia pilosa Ldb of different of vari-ous concentrations affect the immune system of the mice immunosuppressed with cyclophosphamide (CP). Methods BALB/C mice were randomly divided into seven groups, normal control group, model control group, 10, 5, 2.5, 1.25, and 0.625 g/ml medicated groups. The model group and the medicat-ed group were intraperitoneally injected with CP to set up the immunosuppressed model, then the medi-cated group were gavaged with different concentrations of the water decoction of Agrimonia pilosa Ldb each day (0.02 ml/g). After5 days treatment, the blood was obtained to measure the white blood cell (WBC) count and the spleen was dissected to calculate the spleen indices, to investigate how the water decoction of Agrimonia pilosa Ldb of different concentration affect the immune function of the mice. The data was analyzed using SPSS13.0. The differences among multiple groups were assessed using one way ANOVA. The difference between two groups was assessed using Student’s t-test.Results Compared to the normal control group, the model control group had a sharply decreased spleen indexand WBC count (P<0.01). Compared with the model control group, the group of 10 g/ml medicated had a significantly raised WBC count (P<0.05), and as well as all other medicated group (P<0.01). The spleen indices of the medicated group were also significantly higher than the model control groups (P<0.01). The effects in the medicated groups with 2.5 g/ml, 1.25 g/ml, 0.625 g/ml concentrations were significantly higher than the medicated groups of10 g/ml and 5 g/ml for the spleen indices and WBC count.Conclusions The water decoction of Agrimonia pilosa Ldb can dramatically improve the immune function of mice immunosuppressed with CP. The immune function is enhanced with the de-clines of medicine concentration. The results showed that the clinical use of Agrimonia pilosa Ldb has its unique dose response relationship.
