目的 探讨职业砷接触工人miRNA生成和成熟相关mRNA表达变化规律,深入认识砷的致癌机制。方 法 整群随机抽取正在进行砷冶炼的工人43名作为高暴露组,已经停止砷冶炼85d的工人36名作为低暴露 组,对照组24人。用实时荧光定量PCR(RT-PCR)检测外周血miRNA生成和成熟相关mRNA表达情况,采 用氢化物发生原子吸收分光光度法检测尿砷含量。尿砷浓度和相关mRNA的-△△Ct值组间比较采用t检 验。检验水准α=0.05。结果 高暴露组尿砷为(1738.62±1361.62)μg/g肌酐,低暴露组为(561.20±451.50)μg/ g肌酐,对照组为(17.63±11.07)μg/g肌酐,各组间差异具有统计学意义(P<0.01)。高暴露组外周血miRNA生 成和成熟相关mRNA Lin28、Lin28b、Exportin-5、Trbp、Dicer、Ago2 RT-PCR扩增-△△Ct与低暴露组和对 照组间差异均具有统计学意义(P<0.01),而低暴露组与对照组间差异均无统计学意义(P>0.05)。结论 砷冶 炼工人机体内可能存在大量miRNA生成和成熟相关mRNA表达增加。此过程是可逆的,砷中毒患者早诊、 早治非常重要。
Abstract:
Objective To investigate the rule of mRNA expression related with generation and matura-tion of miRNA in workers exposed to arsenic, study the carcinogenic mechanism of arsenic deeply. Methods We selected43 workers in one arsenic plant which was producing arsenic trioxide (high expo-sure group), and 36 workers from another plant which the production had stopped85 days (low expo-sure group) by random cluster sampling. RT-PCR was performed to detect the expressions of mRNAs related with generation and maturation of miRNA. It was student's t test used to analyse the differences along groups for urinary arsenic and the -△△Ct value of relative miRNAs. Test level wasα=0.05. Results The concentration of urinary arsenic were(1738.62±1361.62)μg/g Cr in high exposed group, (561.20±451.50)μg/g Cr in low exposed group, and(17.63±11.07)μg/g Cr in control group, and there were significant difference along three group(P<0.01). The -△△Ct came from RT-PCR for Lin28, Lin28b,Exportin-5, Trbp, Dicer and Ago2 were(9.88±2.42),(8.49±3.10),(9.57±2.36),(6.07±1.62), (6.96±1.60)and(6.91±2.02)respectively in mRNAs(P<0.01), but not between low exposed group and control group for all mRNAs(P>0.05). Conclusions There are many miRNAs which are generating and maturating in workers produce arsenic trioxide. At the same time, the expressions of many relativemRNA increase. The process is reversible, so it is of great important to diagnosis and treat ear-ly patients.
