摘要:
目的通过检测氯乙烯(VCM)对大鼠肝脏细胞P16、CCND1和CDK4基因mRNA表达的变化,从细胞周期调控角度,探讨VCM可能的致癌机制。方法选用健康SD大鼠32只,按体重随机分为4组:阴性对照组,5、25、125 mg/kg氯乙烯染毒组。采用腹腔注射染毒,隔日染毒,每周3次,连续6周。采用实时荧光定量PCR方法,测定大鼠肝脏细胞P16、CCND1和CDK4基因mRNA表达量。结果氯乙烯对大鼠肝脏细胞CCND1和CDK4基因mRNA表达量随着染毒剂量的升高而降低,且125 mg/kg染毒组与对照组比较,差异均有统计学意义(P<005)。P16 基因mRNA表达量没有明显改变,差异无统计学意义。结论VCM对大鼠肝脏细胞周期的影响可能是通过下调CCND1和CDK4基因mRNA的表达。
Abstract:
Objective Detect the change of P16,CCND1 and CDK4 gene mRNA expression in rat liver cells,from the field of cell cycle regulation,to explore the possible carcinogenic mechanism. Methods Thirty-two Sprague Dawley(SD)rats(half male and half female)were randomly divided into negative control group and three experimental groups exposed to 5,25,and 125mg/kg VCM.The rats were treated with clean air and different doses VCM by intraperitoneal injection with three times a week for 6 weeks,and rats were killed at the end of 6 weeks.Real-time quantitative PCR method was applied to detect the change of P16,CCND1 and CDK4 gene mRNA expression in liver cells. Results Compared with the control,CCND1 and CDK4 gene mRNA expression in liver cells of 125 mg/kg dose group decreased significantly(P<0.05).P16 gene mRNA expression did not change significantly. Conclusions VCM affects cell cycle through decreasing CCND1 and CDK4 gene mRNA expression.
